Abstract Introduction and Objective: High-Intensity Focused Ultrasound (HIFU) is an FDA-approved procedure for the treatment of localized prostate cancer. HIFU destroys tumor tissue by noninvasively delivering ultrasonic energy, which rapidly heats targeted tissue. Prostate cancer has few tumor-infiltrating lymphocytes, are required antitumor immunity. The objective this study was to determine if activates immunity increases lymphocyte infiltration into tumor. Methods: RM1 tumors were grown on flanks syngeneic C57BL/6J mice one treated with HIFU. Contralateral growth monitored, collected at end assess immune cell infiltration. Tumor infiltrating lymphocytes flow cytometry. Results: When HIFU, contralateral (untreated) decreased when same type but not it a different type. increased CD4 +and CD8 +lymphocyte Depletion studies confirmed that effect dependent. Conclusions: In preclinical models, generates response capable suppressing mouse tumors.
